Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) is widely used for rapid and high-throughput analysis of biomolecules, including proteins and lipids. However, organic matrices often produce intense background signals in the low-molecular-weight region through fragmentation during laser irradiation, limiting small-molecule detection and interpretation. To overcome this limitation, surface-assisted laser desorption/ionization time-of-flight mass spectrometry (SALDI-TOF MS) has been investigated as an alternative. Among SALDI matrices, TiO₂ has attracted attention because its optical absorption and photocatalytic properties can promote efficient analyte desorption and ionization. In this study, synthesized TiO₂ nanostructures generated lower background signals in the low-molecular-weight region than organic matrices. They also exhibited soft ionization characteristics comparable to P25 and stable signal reproducibility in repeated measurements. Furthermore, the spatial distributions of endogenous and exogenous candidate ions in fingerprint samples were visualized using SALDI-TOF MSI. These findings demonstrate the potential of synthesized TiO₂ nanostructures as inorganic matrices for reproducible low-molecular-weight SALDI-TOF MS and MSI. |
