Patient-derived organoids serve as robust models that closely reflect the histological and molecular characteristics of native human tissues. In this study, to identify key protein markers associated with lung cancer, we performed quantitative proteomics profiling using nanoLC-MS/MS (timsTOF Pro) on pairs of patient-derived lung cancer organoids and normal lung organoids. Proteomics analysis revealed clear differences in the expression patterns of cell adhesion and cytoskeletal complex proteins between tumor and normal organoids. Tumor organoids exhibited a significant increase in cancer-associated epithelial markers and integrin-related proteins (e.g., EPCAM, MUC1, CEACAM5, AGR2, CLDN4, LGALS3, ITGA3, ITGB4, FLNA, SFN, and GSN). In contrast, basement membrane components (LAMA3, LAMB3, LAMC2, COL7A1), hemidesmosomal proteins (ITGA6, PLEC, FERMT1, DST), and cell-cell junctional proteins (CDH1, TJP1, PKP1, PKP3, DSG3, DSC3) were markedly decreased. These results indicate that lung cancer progression is accompanied by a loss of cell-cell adhesion and a concurrent increase in specific cancer-surface markers, highlighting their potential as diagnostic biomarkers and therapeutic targets. |
