Free pantothenic acid in candidate certified reference material was accurately measured with the high-order method based on isotope dilution-liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS). After extraction with 10 mM ammonium acetate (pH 5) and clean-up using protein precipitation, the sample was analyzed using LC-MS. Commercially available beef meat was freeze-dried, pulverized, sieved, and homogenized for the production of the candidate reference material. Then, the processed material was bottled in 15 g portion. The prepared candidate reference material was sufficiently homogeneous as among-bottle relative standard deviation was 0.64% for pantothenic acid. The stability of pantothenic acid was evaluated under -70 ℃, -20 ℃, 4 ℃, and room temperature for up to 12 months, 6 months, 1 months, and 2 weeks, respectively. The content of pantothenic acid was stable throughout each condition. The concentration of free pantothenic acid in the candidate certified reference material was 13.15 ± 0.34 mg/kg. |
