The rapid and efficient analysis of various doping agents—including anabolic-androgenic steroids (AAS), stimulants, and diuretics—is essential in anti-doping testing. We developed a screening method for the simultaneous extraction and detection of the 12 doping agents in human urine using liquid chromatography–tandem mass spectrometry (LC–MS/MS) as a single analytical platform. A 0.5 mL
urine was mixed 0.5 mL of phosphate buffer (pH 7.4) and 20 μL of
β-glucuronidase for hydrolysis. After incubation for 1 h at 55℃, a two-step
liquid-liquid extraction at pH 10 followed by pH 4 were sequentially extracting
with an organic solvent to isolate different components based on their
pH-dependent solubility. The combined organic layer was evaporated to dryness.
The residue was reconstituted in 0.2 mL of 50% methanol, and then injected onto
the LC-MS/MS system. |
