Chloramphenicol (CAP) is an antibiotic that was formerly used in livestock due to its cost-effectiveness, high bioavailability, and potent antimicrobial activity. However, It has been banned in many countries because of its severe adverse effects on human health, including aplastic anemia, leukopenia. Accordingly, the Ministry of Food and Drug Safety (MFDS) of Korea designates CAP as one of 28 substances prohibited in food. As part of Korea’s National Residue Program (NRP), regular monitoring is conducted for CAP residues in major livestock products. The Korean Food Code currently sets the limit of quantification (LOQ) for CAP at 0.5 µg/kg. To ensure compliance with international standards, especially for samgyetang exports to the EU, an analytical method using LC-MS/MS was validated for the detection of CAP in chicken muscle, in accordance with Codex Guidelines (CAC/GL 71-2009). The method exhibited LOQ of 0.15 µg/kg, high accuracies within 2.47% and relative standard deviations (RSDs) of 4.82% and 3.93% for intraday and interday precision, respectively. The validated method satisfies the stringent sensitivity requirements outlined in European Commission Regulation 2019/1871, which designates the Reference Point for Action (RPA) for CAP at 0.15 µg/kg. The results demonstrate that the method is highly effective for monitoring CAP residues at extremely low concentrations and is fully suitable for ensuring regulatory compliance in the context of international trade. |
