Glioblastoma is an aggressive form of brain with little therapeutic options available, and correspondingly the 5-year survival rate remains <10% and the median at 15 months. Of particular interest in disease progression is the activation of specific proteins via phosphorylation by protein kinases. Trialling of new therapies therefore may wish to profile these pathways through measurement of total activation and phosphorylation of specific serine/threonine residues, and also kinase abundance. Multi-reflecting time-of-flight mass spectrometry with parts-per- billion mass accuracy coupled to the selectivity of TofMRM provides an avenue to accurately and definitively characterize the level of post-translational modifications while also providing quantitative data surrounding the identified peptide markers. Marker peptides found in glioblastoma cell lines either untreated or treated with a metabolic therapy were subject to absolute quantitation using heavy isotopically labelled peptides, and their abundance compared between groups.
