A
highly accurate method based on isotope dilution-liquid chromatography/tandem
mass spectrometry (ID-LC/MS/MS) has been developed for the determination of
vitamin B12 in beef. Isotope labeled cyanocobalamin-[13C7]
analogue of the target analyte was used as an internal standard. For sample
preparation, various forms of vitamin B12 existing in beef were
released and converted to cyanocobalamin, the most stable form of vitamin B12,
by protease treatment using papain and cyanide with 0.01 M phosphate-buffered
saline (pH 7.2) as an extraction solvent. The supernatant of the sample extract
was purified with an immunoaffinity column (EASI-EXTRACT VITAMIN B12,
IAC). LC system connected to an Acquity HSS T3 C18 column was used with
gradient elution to separate the 4 major forms of vitamin B12;
cyanocobalamin, hydroxocobalamin, methylcobalamin, adenosylcobalamin from
matrix interferences. The matrix effect profile identified by the post-column
infusion system showed that the selected sample preparation procedure using the
IAC is efficient for the elimination of co-extracted matrix interferences. The
developed ID-LC-MS/MS method showed sufficient accuracy (97.1~102.6 %) with a
relative expanded uncertainty of 1.76 ~ 4.36 %. Intra-day and inter-day
relative standard deviations were less than 2% and 1%, respectively. Therefore,
this method demonstrated adequate metrological quality and can be used for the
value-assignment of beef-certified reference materials.
